Presentation Topic: Non-Small Cell Lung Cancer (NSCLC) Treatment and PROTAC as a New Targeted Therapy Strategy
Lung cancer is a leading cause of cancer-related deaths, and non-small cell lung cancer (NSCLC) makes up 85% of all cases. A subset of NSCLC patients harboring epidermal growth factor receptor (EGFR) mutations can be treated with EGFR targeted kinase inhibitors, but resistance will ultimately occur and new strategies that overcome drug resistance are urgently needed. In addition to immunotherapy, bispecific antibody, and antibody-drug conjugate, proteolysis-targeting chimera (PROTAC) has emerged as a paradigm-shifting approach to address the resistance mutation issue. In this presentation, the discovery and development of a potent, selective and orally bioavailable EGFR-PROTAC molecule HJ-004 will be presented and discussed. This approach can be extended to other actionable mutations in NSCLC such as KRAS, ALK, ROS1, and BRAF in order to improve patient treatment outcomes.
