Short Biography
Dr. Hai-Bing Zhou is currently a professor in the Wuhan University School of Pharmaceutical Sciences, director of the Hubei Province Engineering and Technology Research Center for Fluorinated Pharmaceuticals, PI of State Key Laboratory of Virology and Biosafety. His research interests include the innovative drug discovery based on the structure and mechanism of action of biological targets, with a focus on diversity-oriented small molecule inhibitors and targeted protein degradation (TPD) agents. He also serves as the member of the Medicinal Chemistry Professional Committee of the Chinese Pharmaceutical association, editorial board member for multiple journals, including Journal of Medicinal Chemistry (2022-2024), Acta Pharmaceutica Sinica, and Pharmaceutics etc. He has published more than 100 papers and holds more than 30 patents.
Presentation Topic: Discovery of Novel PROTAC Degraders Targeting the Protein-Protein Interaction of Estrogen Receptor and Coactivator
• Targeted protein degradation (TPD) provides a new drug intervention approach for protein-protein interaction (PPI).
• The oncogene steroid receptor coactivator-3 (SRC-3) can directly participate in the transcriptional regulation mediated by estrogen receptor (ER) to promote the occurrence and progression of endocrine resistance in breast cancer.
• Novel PROTAC degraders targeting the ERα and SRC-3 interaction were designed and synthesized, which displayed excellent anti-breast cancer activity in drug-resistant xenografted models.
• This proof-of-concept study may offer a novel degradation strategy to overcome endocrine resistance, providing new opportunities for the development of innovative treatments for ERα+ breast cancer.
